The main objective of the 3D study is to create a large pregnancy and birth cohort, allowing for the assessment of the effects of combined prenatal exposure on both birth outcomes and longer-term infant and child health.
The specific objectives are to:
* Determine the effects of pre- and post-natal exposures to environmental contaminants, nutritional factors and psychosocial stress on pregnancy and neurodevelopmental outcomes;
* Assess the contribution of social, genetic, epigenetic, nutritional and environmental factors to the etiology and outcomes of IUGR and preterm birth;
* Assess the effects of assisted reproductive technologies (ART) on indicators of epigenetic modifications, and the relationship between such epigenetic changes and child neurodevelopmental, cardiovascular, and metabolic outcomes; and
* Understand the mechanisms linking children's early development with mental health after the transition to formal school by examining the challenges and available resources during the transition from preschool to school, and evaluating the child’s adjustment to school and his/her new environment (3D-Transition study) .

- Pregnant women and their partners were recruited during the first trimester of pregnancy (8–14 weeks) and followed up at mid (20–24 weeks) and late (32–35 weeks) pregnancy, and at delivery. Postnatal visits took place at 3 months, 1 year, and 2 years following the birth. From age 4 years onward, both parents and children were followed at 4, 5, 6, 7, 8, 9 and 10 years after birth (3D-Transition Study). Starting in 2023, parents and adolescents (cohort 1 aged 12 years, cohort 2 aged 11 years, and cohort 3 aged 10 years in 2023) are being followed annually until 2029 (3D-Transition ADO follow-up).

The population is composed of 8 to 14 weeks pregnant women from the general population between 18 and 47 years of age, who are fluent in French or English, who were attending prenatal clinics (ultrasound, midwife and/or doctor’s clinics) during the first trimester of pregnancy and who plan to deliver in a study hospital.

- Exclusion criteria: Multiple pregnancies, intention to donate or bank cord blood, intravenous drug users, or if the woman has any one or more of the following conditions: HIV+ status, renal disease with altered renal function, any collagen vascular disease requiring active treatment (e.g. lupus, scleroderma), epilepsy, cardiovascular disease, serious pulmonary disease, cancer, or severe hematologic disorder.
